3D-Marfan Smooth Muscle Morphology

What you are going to do
Marfan Syndrome patients have a mutation in fibrillin-1 (FBN1), leading to skeletal, ocular and severe cardiovascular problems. One of these cardiovascular problems is widening of the aorta (aneurysm) with aortic rupture as main risk factor with often fatal consequences. These aortic problems may already occur at a very young age and patients usually die before the age of 40 if they are not operated for aortic replacement in time. We aim to prevent or postpone aortic surgery by identification of pharmacologic treatment strategies. However, over 2000 different mutations have been described in the FBN1 gene causing Marfan Syndrome. So far, we identified two categories of Marfan patients, based on the type of mutation, explaining disease severity and drug sensitivity. In the current project the PhD candidate will collaborate with the 3D-cell culture/imaging company OcellO. Vascular smooth muscle cell 3D-culture assays will be developed to understand the impact of different FBN1-mutations on smooth muscle cell morphology and intracellular signaling pathways. We aim to stratify patients based on these characteristics and predict their drug sensitivity. This study also involves close collaboration with the physicians treating Marfan patients.

 What we expect from you
A recent MSc in Medical/Molecular Biology or related discipline, preferably with a background in molecular cell biology and pathology. We expect a team player with a drive for science, highly motivated to become an independent researcher.

Where you are going to work
The PhD candidate will work in the group of Dr. de Waard at the department of Medical Biochemistry in the AMC. He/she will work in a team of experienced staff, PhD candidates and technicians with excellent infrastructure. There will be intense collaboration with OcellO and the Marfan clinic (Cardiology/Genetics) at the Amsterdam UMC.

  • The maximum gross monthly salary based on a 36-hour work week is € 2.992,- (scale OIO CAO UMC).commensurate with qualifications and experience
  • The base salary does not include holiday pay (8%) and a year-end bonus (8.3%).
